Parse Evercode

The Parse Biosciences Evercode™ split-pool combinatorial barcoding system involves multiple rounds of barcoding of fixed and permeabilised cells.
Kits from Parse Bioscience are available in several versions for variable sample sizes and numbers, ranging from the Evercode WT mini for 1-12 samples targeting a total of 10,000 cells to the Evercode WT Penta which is available in versions of 1-384 samples and up to 5 million cells.
Evercode is a whole transcriptome, RT-based method using both poly-dT and random hexamer priming.
The Evercode system works with fixed cells or nuclei.

Whole transcriptome (WT)
Parse Biosciences currently offers the whole transcriptome WT Mini, WT, and WT Mega kits, to enable single cell sequencing at a scale to suit any project.
Immune Receptor analysis (TCR/BCR)
Parse Biosciences currently offers the Evercode TCR and Evercode BCR protocols with combined whole transcriptome and immune receptor profiling (human & mouse).
Targeted sequencing
Parse currently offer GeneSelect targeted single-cell sequencing – choose from the Immune1000 panel or design your own.
CRISPR gene perturbations
Use Parse CRISPR Detect to study gene perturbations in your CRISPR studies & look at target validation in eg. drug discovery, differences in treatment responses, and mapping of pathways involved in cell differentiation.
Sample preparation
In the Parse Evercode workflow the cells/nuclei are first fixed. Fixed samples can be stored at -80C, resulting in flexibility in analysis. Samples must be fixed with Evercode fixation kits.
Sample Compatibility
The technology is species agnostic (except for the TCR/BCR profiling) and reverse transcription is performed using both a poly-dT primer and random hexamers.
